Which Transformer toy with no added parts is the biggest size shifter from one mode to another. My nephew just open CV Breakdown & stated how much small van mode is to robot mode. I know he isnt the biggest but is an example.

Excellent question. It's got to be one of the Japanese shellformers, doens't it? BWN Dead End, maybe? He has a robot mode that folds out and carries his altmode.

That is a good question. Not sure what the answer is though. Errrr.... G1 Brawn made a fairly tall robot... Ooh what about that outstanding Movie Lockjaw figure? His robot mode is as tall as a voyager.

Animated Lockdown went from a normal sized car to a pretty giant chunky robot

BWN Dead End (despite giving Shellformers a whole new ironic meaning) is one of my faves, because ammonite. But he really doesnt gain or lose any size between modes.
Cybertron Primus/Cybertron "shrank" considerably, in planet mode. If I recall, Cyb Hotshot seemed to pack a fairly impressive robot in an average car.
Deathstar Vader did too, though not an actual TF .
Animated Lockdown was a voyager equivalent robot in a deluxe car body. Animated Ratchet, not quite as much, feels pretty compact.
I'd have to say ALL G1 cassette TFs. Those were pretty amazing engineering. Even now. G1 Brawn/Outback had a nifty unfolding trick that made him feel more impressive in robot.
There are others, but unfortunately I'm blanking ATM.

Animated & Movie Lockdown are the ones that spring immediately to mind. Prime RID Vehicon does some impressive folding though I have a feeling that has the effect of reducing kibble more than increasing size.
Big Convoy certainly spreads out a lot.

The first figure I thought of was Universe Onslaught- a relatively small APC that turned into a towering robot that was as tall as some leader-class figures. Then I remembered the original Skids. An awful lot of robot packed into a very small car.

Good call on Skids, Nickos.
And good call on the G1 cassettes, CJ. Masterpiece Laserbeak and Buzzsaw seem to be more hefty than their MP cassetes chumbs.

G1 Optimus Prime - When he turns from alt-mode truck to a robot, the trailer mass just *disappears*! (At least that's what the cartoon taught me)

When I first read the question I thought we were talking about the fictional 'mass shifting' flange some Transformers use i.e. Megatron...
I immediately thought of the Death Star and MP tapes too. Some of the other Star Wars Crossovers are impressively compacted too such as the Luke Snow Speeder (LOVE that toy!) and Millenium Falcon (splits into two full size robots). Boulder and some of the other Rock Lords used to squash down quite small compared to other toys of the time. Toyworld's Bii does really well to fold up into a Penny Racer-style Bug. Recently, I'd say FoC Soundwave and Blaster turn out larger than their alt mode would have you think.
On aggregate though, the mini Autobots probably win as a series as nearly all of them relied on parts 'folding out' to the point where their dimensions are noticeably increased for their robot form.
